W-Muricholic Acid (Omega-Muricholic Acid / Omega-Muricholat) is a human metabolite, a member of the class of muricholic acids.
Muricholic acids are of the group of primary bile acids (BAs) found predominantly in mice (where their name comes from) and detectable at low levels in other species. However, muricholic acids are also present in the urine of healthy humans, especially neonates.
Muricholic acids are different from the other primary BAs in the human body - cholic acid and chenodeoxycholic acid (CDCA), because they contain a hydroxyl group at positions 6 and 7, and can have α- or ß-configuration, respectively. The hydroxylation reactions that generate muricholic acids in rodents happen under the regulation of the cytochrome P450 Cyp2c70 enzymes, and muricholates are obtained as a result.
BAs are distinct naturally-occurring molecules with many unique properties. They derive from cholesterol in the liver through a multistep process, involving a range of specific enzymes. They undergo an enterohepatic circuit and are crucial for the digestion, absorption, and overall metabolism of fats, sterols, fat-soluble vitamins, hydrophobic nutrients, and more. BAs are unique in their properties since they also function as signaling molecules that not only modulate their own biosynthesis and homeostasis, but participate in the control of key metabolic pathways, involving cholesterol, glucose, energy, and drug metabolism, as well.
As with all BAs, muricholic acids, including W-Muricholic Acid, have been proven to be potent regulators of BA synthesis, bile flow, and BA excretion and studies reveal a strong positive feedback regulating mechanism.
Scientists have also recently suggested that treatment with muricholic acids, alongside W-Muricholic Acid, is beneficial against metabolic syndrome. The BAs have been found to manifest increased resistance against weight gain, glucose intolerance, and steatosis, thanks to their abilities to decrease absorption of cholesterol.
